Expression of (1-3)-B-Glucanases in Developing and Germinating Barley Kemels

@EN Expression of (1-3)-¥â-glucanases was examined in germinating and developing barley kernels. (1-3)-¥â-glucanase activity detected in kemels land developing roosts and shoots was mostly contributed by three major isoforms, one acidic and two
basic
isoforms designated A, BI and BII, repectively. Drastic increase in enzyme activity during germination was due to the subsequent increase in the expression of the three major isoforms, especially of the isoform Bi. The enzyme activity in
developing
kernels was much lower than in germinating kernels, and the activity decreased rapidly as kernels matured. The lower levels of the enzyme activity in developing kernels were due to the lower expression of all three major isoforms. Tissue specific
expression of (1-3)-¥â-glucanase isoforms was also examined in 5-day old seedlings. Relatively high levels of enzyme activity were detected in roots and shoots as well as in kernels. (1-3)-¥â-glucanase activity in kernels 5 days after gemination
was
mostly attributed to the action of the basic isoform BII and the acidic isofrom A. Contribution of the basic isoform BI was relatively low in kernels. In contrast, (1-3)-¥â-glucanase activities detected in roots and shoots of 5-day old seedlings
were
mostly contributed by the basic isoform BI. Contribution of the basic isoform BII was very low in roots and shoots. Expression of the acidic isoform A was lowest in roots. These results indicate that most of the (1-3)-¥â-glucanase activity in
barley is
attributed to the temporal and spatial expression of the three major isoforms. Expression of (1-3)-¥â-glucanases in healthy developing tissues suggests that in addition to possible role in defense, (1-3)-¥â-glucanases may have a complex
association
with
normal developmental processes in barley plants.
